dc.description.abstractIntroduction & Objective: Ulcerative colitis is a chronic inflammatory disorder involving the large intestine and rectum. However, the current therapy available is with limited efficacy and accompanied side effects. Thus, this study was conducted with the following objectives: 1. To determine the effect of Mucuna pruriens seed extract in Tri Nitro Benzene Sulfonic Acid (TNBS) model of experimental colitis in male Wistar rats. 2. To determine the effect of Mucuna pruriens seed extract on levels of inflammatory cytokines (TNF-α, IL-1β, IL-6) Methods: Experimental colitis was induced in Wistar rats using Tri-nitro benzene sulfonic acid (TNBS). Animals were divided into 4 groups with n=8 in each group. Induction of colitis was done using a modified infant feeding tube (6F) inserted per-rectally, under anaesthesia. Treatment was given with standard prednisolone (4.5mg/kg), test drugs, Mucuna pruriens (400mg /kg) once a day for 14 days. Weight measurement, food intake and Disease Activity Index was measured during the study period. At the end of study period rats were sacrificed and colon was dissected out. Half colon was used for gene expression study for TNF- α, IL-6 and IL-1b. Blood was collected for measuring oxidative stress. Remaining colon tissue was fixed in 10% formalin and sent for histopathological studies.en_US
