Abstract: ABSTRACT Background Helminths parasites are considered as a master regulator for the immunomodulation. Macrophage migration inhibitory factor-2 is one among of the homologue’s molecules of the host, secreted by the W. bancrofti. It is a pleotropic cytokine secreted by different immune cells and plays a significant role in immune mediated disorder. Interestingly Human MIF interacts with insulin and helps in its proper folding and release from the pancreatic β-cells. Immuno-metabolic diseases are the most demanding arena in the medical sciences is to develop new biological molecules. Parasitic Wba-MIF2 could be one promising molecule for the immune mediated diseases. Our aim of the study was to check the immunomodulatory potential of the Wba-MIF2 in diabetic animal model, phosphoproteomics signaling in macrophage cell line and Wba-MIF2-Insulin interating peptide identification. Objectives Objectives of the study are a) Expression and purification of the Wba-MIF2. b) To investigate the immunomodulatory effect of Wba-MIF 2 in diabetic rats.c) To understand the role of Wba-MIF2 in downstream signaling of immune cells using LCMS/ MS based phospho-proteomics approach. d) To identify the Wba-MIF2 epitopes using bioinformatic tools and its in-vitro testing for insulin oxidoreductase activity.
